Up until now, freezing and testing of PBMC has required the use of serum. Serum contains a plethora of bioactive molecules whose variable concentrations render each batch unique. Each lot must therefore be tested, which makes working with serum both expensive and time-consuming.

PBMC of different healthy donors (A-C) were tested in an IFN-γ ELISPOT assay directly ex vivo ('Fresh') and after cryopreservation with CTL freezing media ('Frozen'). The mean number of IFN-γ spots induced by the specified antigens is shown for the fresh and the frozen PBMC. In addition, the viability and recovery rate of the thawed cells is shown.
