ImmunoSpot® software permits accurate ELISPOT counting at single cell resolution validated experimentally (Refs. 1, 2). By eliminating subjectivity from the counting and by creating complete transparency of the counting results, ImmunoSpot® software elevates ELISPOT analysis to an exact science.
Transparent and Accurate ELISPOT AnalysisAs the first step of ELISPOT analysis/documentation, ImmunoSpot® Series 3 Analyzers save tamper proof, 21 CFR Part 11 compliant image files ("photographs") of ELISPOT wells (the images on the left). The saved high resolution image files provide a permanent record of the assay results long after the original plates have decayed, permitting to review, and to reanalyze the data. In the second step of ELISPOT analysis, the ImmunoSpot® Software fully automatically counts spots on any number of plates. User definable, but constant counting parameters are used for analyzing all wells within an assay. This procedure permits the objective comparison of assay results in different wells/plates. Following GLP guidelines, overlays of the raw image files and of the counting results are saved for each well (the image in the middle), as are the counting parameters. The results of the automated counting therefore become transparent, documented, and can be easily reproduced. In the third step of ELISPOT analysis, the results of the automated counting need to be quality controlled. Corrections need to be made if a well contains clusters (as shown it the example well provided, and as frequently occurs when freeze-thawed PBMC are tested). In the quality control step, the cluster can be manually excised. The software will then recalculate the spot numbers assuming that the spot size and density would be the same in the excised area as it is in the rest of the well (the images on the right). The ImmunoSpot® software saves and annotates such changes. Versatile Data Management!ImmunoSpot® software automatically organizes your ELISPOT analysis by managing the raw data, the counting and quality control results. The software permits to "bundle" plates, so that all plates within an experiment can be conveniently counted, quality controlled and managed in a batch. CTL's accessory SpotMap™ Software permits to further maximize data management by including information on test subjects, antigens, cell numbers, and other parameters of the assay. Presentation FeaturesImmunoSpot® permits viewing, printing, exporting image files at selectable resolutions, and zooming in on well segments of specific interest.
